SKLRD Tianyu Zhang’s Research Team Established the Selectable Marker-free SfAlPa for Rapid, Serial, Quantification of Pseudomonas aeruginosa in Live Mice with One-step Method

Recently SKLRD PI’s Researcher Tianyu Zhang’s research team combined the autonomous luminous operon element, Mini-Tn7 system and Xer/dif system based on biological principles and techniques with endogenous XerC/D enzyme-removing resistance genes, successfully establishing the selectable marker-free SfAlPa with one-step method. The research finding titled “Rapid, serial, non-invasive quantification of Pseudomonas aeruginosa in live mice with a selectable marker-free autoluminescent strain” was published on the international academic periodical Biosensors and Bioelectronics (IF:10.257) online.

The selectable marker-free SfAlPa bacterial colonies established by the research team can not only glow turquoise in the dark room, but also remarkably allow the researchers to see the infected mice glowing and take sharply focused photos with a home camera. The light unit and bacterial load correlate well to each other, and the glowing is quite stable. The mouse infection model established with SfAlPa can realize non-invasive, efficient and real-time monitoring of bacterial load in live mice and its distribution change, prominently enhance the screening efficiency of anti-Pseudomonas aeruginosa drugs, and sharply reduce the number of mice needed and economic cost.
